Volasertib (compound code BI 6727) is an investigational, potent, and selective inhibitor in vitro of PLK1, an important regulator of the cell cycle. Preclinical studies show that BI 6727 is a specific PLK1 inhibitor. As observed in treated cancer cells, BI 6727 may be associated with disruption of the mitotic spindle, spindle checkpoint activation, and prolonged mitotic arrest in prometaphase (“Polo arrest”), which subsequently leads to cell death by apoptosis.
