Boehringer Ingelheim's next generation tyrosine kinase inhibitor under investigation. Afatinib (compound code BIBW 2992) is an irreversible inhibitor of both epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2) kinases. Both of these are involved in cell proliferation, differentiation and programmed cell death (apoptosis) and inhibition may play a critical role in the prevention of tumour growth and spread. Afatinib (compound code BIBW 2992) is currently being investigated in advanced non-small cell lung cancer (NSCLC), breast cancer, head and neck cancer, colorectal cancer and glioblastoma. Its safety and efficacy have not yet been fully established.
